Understanding Loss on Drying (LOD)
Loss on Drying (LOD) is a standard test method (e.g., USP <731>) used to determine the amount of volatile matter (usually water) in a pharmaceutical sample. It typically involves heating the sample below its melting point in an oven until it reaches a constant weight.
Step 1: Weigh Initial Sample
Weigh the sample container (tare), add the sample, and record the Initial Wet Weight. Ensure the balance is calibrated.
Step 2: Dry the Sample
Place the sample in a drying oven or moisture analyzer at the specified temperature (e.g., 105°C) for the required time period.
Step 3: Weigh Final Sample
After cooling in a desiccator, weigh the sample again to obtain the Final Dry Weight. Enter both values above to calculate the percentage loss.
